Stereology is a valuable tool for neuroscientists, allowing them to obtain 3-Dimensional information from 2-Dimensional measurements made on appropriately sampled sections. This 3-D information is invaluable in correlating structural/functional relationships in the pursuit of far greater understanding of the function of the central nervous system. However, in carrying out such measurements, often based on limited data sets, there is a risk of experimenter bias. An important feature of modern design based stereology is to be aware of potential sources of bias (...) and eliminate them during the data collection. With many of the major neuroscience journals now insisting that quantitative data be presented, there is a greater need than ever for neuroscientists to understand the theory and practice behind quantitative methods, such as those offered by stereology. Quantitative Methods in Neuroscience is a cookbook of stereological methods written especially for neuroscientists. It provides clear and accessible advice about when and when not to use stereology. Airway mucin secretion is important pathophysiologically and as a model of polarized epithelial regulated exocytosis. We find the trafficking protein, SNAP23, selectively expressed in secretory cells compared with ciliated and basal cells of airway epithelium by immunohistochemistry and FACS, suggesting that SNAP23 functions in regulated but not constitutive epithelial secretion. Heterozygous SNAP23 deletant mutant mice show spontaneous accumulation of intracellular mucin, indicating a defect in baseline secretion. However mucins are released from perfused tracheas of mutant and wild-type mice at the (...) same rate, suggesting that increased intracellular stores balance reduced release efficiency to yield a fully compensated baseline steady state. In contrast, acute stimulated release of intracellular mucin from mutant mice is impaired whether measured by a static imaging assay 5 min after exposure to the secretagogue ATP or by kinetic analysis of mucins released from perfused tracheas during the first 10 min of ATP exposure. Together, these data indicate that increased intracellular stores cannot fully compensate for the defect in release efficiency during intense stimulation. The lungs of mutant mice develop normally and clear bacteria and instilled polystyrene beads comparable to WT mice, consistent with these functions depending on baseline secretion that is fully compensated. This paper develops the outlines of a pragmatic, adaptive management-based approach toward the control of invasive nonnative species (INS) through a case study of Kings Bay/crystal River, a large artesian springs ecosystem that is one of Florida’s most important habitats for endangered West Indian manatees (Trichechus manatus). Building upon recent critiques of invasion biology, principles of adaptive management, and our own interview and participant–observer research, we argue that this case study represents an example in which rigid application of invasion biology’s (...) a␣priori imperative to minimize INS has produced counterproductive results from both an ecological and social standpoint. As such, we recommend that INS control in Kings Bay should be relaxed in conjunction with an overall program of adaptive ecosystem management that includes meaningful participation and input from non-institutional stakeholders. However, we also note that adaptive management and INS control are by no means mutually exclusive, in Kings Bay or elsewhere. Instead, we suggest that adaptive management offers a means by which INS control efforts can emerge from—and be evaluated through—ongoing scientific research and participatory dialogue about the condition of specific places, rather than non-contextual assumptions about the harmfulness of INS as a general class. This chapter reviews studies of sex differences in drug abuse. There is increasing preclinical and clinical evidence that gonadal hormones play a role in the subjective effects, reinforcing effects, and other effects of abused drugs. Despite this growing evidence, most studies that include women ignore the menstrual cycle. Until more studies are conducted to provide sufficient evidence that the subjective effects of a particular drug do not vary across the menstrual cycle, studies comparing males and females need to control for (...) menstrual cycle phase in a systematic fashion. In recent analytic philosophy, there is a growing interest in the project of conceptual engineering. This paper examines two ways this project might be applied to scientific research, specifically anthropological research. It argues that both of them are harmful to this research. Specifically, it argues that a reliance on the axiological standards of analytic philosophy conflicts with the goal of empirical adequacy. Section one proffers two forms that the engineering project might take when applied to the science. Section two proffers (...) a case study drawn from anthropology. Specifically, it lays out Evans-Pritchard’s Witchcraft, Oracles, and Magic Among the Azande, paying particular attention to the inconsistency he discovered in Azande witchcraft. Sections three and four argue that attempting to remove this inconsistency via the two forms of engineering disable critical features of emic research and discard empirical findings. Finally, I close by noting how the conflict between axiological standards and empirical adequacy might generalize beyond anthropology. (shrink)

In “Understanding Demonstratives”, Gareth Evans bites the bullet regarding Rip van Winkle cases in cognitive dynamics: the fact that Rip sleeps for twenty years and completely loses track of time means he is unable to retain his original belief that “Today is a fine day”. In this paper, the author argues that Evans need not bite this bullet because there are resources in his account of the cognitive dynamics involved in belief retention developed in The Varieties of Reference (...) to successfully confront the challenge posed by the Rip van Winkle case. In particular, when we combine the two central elements of Evans’s cognitive dynamics – the skill of keeping track of one’s spatio-temporal location in addition to memory – it is possible to arrive at the conclusion that it is indeed possible for Rip to retain and re-express his original belief. The extent of tissue resection is determined by the "trade off" between cancer control and the perioperative, functional and aesthetic morbidity and mortality of the surgery. Our recent paper advocating adaptive management of invasive nonnative species (INS) in Kings Bay, Florida received detailed responses from both Daniel Simberloff, a prominent invasion biologist, and Mark Sagoff, a prominent critic of invasion biology. Simberloff offers several significant lines of criticism that compel detailed rebuttals, and, as such, most of this reply is dedicated to this purpose. Ultimately, we find it quite significant that Simberloff, despite his other stated objections to our paper, apparently agrees with our argument that proposals (...) for alternative management of established INS (i.e., alternatives to minimization/eradication) should not be rejected on an a␣priori basis. We argue that more specific development and application of adaptive approaches toward INS management, whether in Kings Bay or other appropriate case studies, would be facilitated if ecosystem managers and invasion biologists follow Simberloff’s lead on this key point. While Sagoff largely shares (and, indeed, served as a primary source for developing) our general arguments that challenge common moral and scientific assumptions associated with invasion biology, he does question our suggestion that participatory adaptive management provides an appropriate framework for approaching environmental problems in which science and politics are inherently entangled. We attempt to answer this criticism through a brief sketch of what participatory adaptive management might look like for Kings Bay and how such an approach would differ from past management approaches. Introduction: Schools have become a primary setting for providing mental health care to youths in the U.S. School-based interventions have proliferated, but their effects on mental health and academic outcomes remain understudied. In this study we will implement and evaluate the effects of a flexible multidiagnostic treatment called Modular Approach to Therapy for Children with Anxiety, Depression, Trauma, or Conduct Problems on students' mental health and academic outcomes.Methods and Analysis: This is an assessor-blind randomized controlled effectiveness trial conducted across five (...) school districts. School clinicians are randomized to either MATCH or usual care treatment conditions. The target sample includes 168 youths referred for mental health services and presenting with elevated symptoms of anxiety, depression, trauma, and/or conduct problems. Clinicians randomly assigned to MATCH or UC treat the youths who are assigned to them through normal school referral procedures. The project will evaluate the effectiveness of MATCH compared to UC on youths' mental health and school related outcomes and assess whether changes in school outcomes are mediated by changes in youth mental health.Ethics and Dissemination: This study was approved by the Harvard University Institutional Review Board. Faced with troubling professional decisions in his long and successful career as a psychiatrist, M. O'C. Drury turned for direction to the philosophical work of his teacher and friend, Ludwig Wittgenstein. Of particular concern to Drury were the situations in which psychiatrists were expected to differentiate between instances of madness that were religious in form and instances of genuine religious experience that, for their oddity, landed believers in psychiatric consulting rooms. In this essay we consider the special orientation Wittgenstein's philosophy (...) gave Drury, for example the way in which Drury came to understand how even his search for a principle of differentiation between madness and religion was misleading and contrary to his own practice—how it involved ‘sitting back in a cool hour and attempting to solve this problem as a pure piece of theory. Using covariant derivatives and the operator definitions of quantum mechanics, gauge invariant Proca and Lehnert equations are derived and the Lorenz condition is eliminated in U(1) invariant electrodynamics. It is shown that the structure of the gauge invariant Lehnert equation is the same in an O(3) invariant theory of electrodynamics.

Evans, c'est la relativité de la vitesse relative, qui (...) n'existe pas en physique classique.The author shows that although Einstein's argument seems classical in almost all points, it fails to be in one very important point: the equality of the velocity of light for all observers. When setting aside this postulate, one returns to the absolute simultaneity. On the other hand, this almost classical argument is perfectly valid from the relativistic point of view and appears as a didactic cleverness. The practice of psychotherapy, or any form of counseling, inevitably requires an anthropological foundation from which to work. Defining the nature of personhood is a task which necessitates an explanation of the various aspects of the self, and the relationships between those aspects. In particular, it is crucial to delineate how the person is both an individual being and a relational being, for we intuitively experience life in both dimensions. These are two characteristics which have often been presented in opposition (...) to each other, however. What is needed is a theory which balances the individual character of personhood with the social character of personhood. The task at hand is to develop such a balance which can serve as a foundation for a Christian psychology. ;C. Stephen Evans, a philosopher and post-empirical critic of modern psychology, has emphasized the individual aspects of personhood. Soren Kierkegaard has influenced Evans' view of the self. Thus, Evans conceives of the person as fundamentally an agent, who chooses freely, acts purposefully and reasonably, and is responsible for the consequences of those choices and actions. ;Ray S. Anderson has developed a theological anthropology and on that basis has articulated a theoretical view of Christian counseling. His theory of the nature of personhood, influenced by the theologies of Karl Barth and Dietrich Bonhoeffer, is a thoroughly social perspective. The person is essentially relational, such that one is social before one is psychological. ;The present work constructs an integrative theory which balances the individual and social insights from Evans' and Anderson's views, respectively. It is accomplished through an analysis of the self across two ontological statuses and three orientations . A biblical paradigm is developed to illustrate the grounding of that theory in a Christian framework. Implications for therapeutic interventions are provided. (shrink)

Aims: The experience of a compromised sense of ‘self’, termed ego-dissolution, is a key feature of the psychedelic experience and acute psychosis. This study aimed to validate the Ego-Dissolution Inventory (EDI), a new 8-item self-report scale designed to measure ego-dissolution. Additionally, we aimed to investigate the specificity of the relationship between psychedelics and ego-dissolution. Method: Sixteen items relating to altered ego-consciousness were included in an internet questionnaire; 8 relating to the experience of ego-dissolution (comprising the EDI), and 8 relating to (...) the antithetical experience of increased self-assuredness. Items were rated using a visual analogue scale. Participants answered the questionnaire for experiences with classical psychedelic drugs, cocaine or alcohol. They also answered the 7 questions from the Mystical Experiences Questionnaire (MEQ) relating to the experience of unity with one’s surroundings. Results: 691 participants completed the questionnaire, providing data for 1828 drug experiences (1043 psychedelics. 377 cocaine. 408 alcohol). Exploratory factor analysis demonstrated that the 8 EDI items loaded exclusively onto a single common factor, which was orthogonal to a second factor comprised of the items relating to increased self-assuredness (rho= -.110), demonstrating discriminant validity. The EDI correlated strongly with our measure of unitive experience (rho =.735), demonstrating convergent validity. EDI internal consistency was excellent (Cronbach’s alpha 0.93). Three analyses confirmed the specificity of ego-dissolution for experiences occasioned by psychedelic drugs. Firstly, EDI score correlated with drug-dose for psychedelic drugs (rho=.371), but not for cocaine (rho=.115) or alcohol (rho=-0.055). Secondly, the linear regression line relating the subjective intensity of the experience to EDI was significantly steeper for psychedelics (unstandardized B coefficient= 0.701) compared with cocaine (0.135) or alcohol (0.144). Finally, a binary support vector machine classifier identified experiences occasioned by psychedelic drugs vs. cocaine or alcohol with over 85% accuracy using ratings of ego-dissolution and ego-inflation alone. Conclusions: Our results demonstrate the psychometric structure, internal consistency and construct validity of the EDI. Moreover, we demonstrate the close relationship between ego-dissolution and the psychedelic experience. The EDI will facilitate the study of the neuronal correlates of ego-dissolution, which is relevant for psychedelic-assisted psychotherapy and our understanding of psychosis. (shrink)

In the past two to three decades health behavior scientists have increasingly emphasized affect-related concepts (including, but not limited to emotion) in their attempts to understand and facilitate change in important health behaviors, such as smoking, eating, physical activity, substance abuse, and sex. This article provides a narrative review of this burgeoning literature, including relevant theory and research on affective response (e.g., hedonic response to eating and drug use), incidental affect (e.g., work-related stress as a determinant of alcohol use), affect (...) processing (e.g., anticipated regret for illicit sex or skipping an exercise session), and affectively charged motivation (e.g., cigarette craving). An integrative dual-processing framework is presented that suggests pathways through which affect-related concepts may interrelate to influence health behavior. (shrink)

The depletion effect, a decreased capacity for self-control following previous acts of self-control, is thought to result from a lack of necessary psychological/physical resources (i.e., “ego depletion”). Kurzban et al. present an alternative explanation for depletion; but based on statistical techniques that evaluate and adjust for publication bias, we question whether depletion is a real phenomenon in need of explanation.
